For access points to discover the controller which is required option 78 (OR) OPTION 43.
Should both be used or one is enough.
-> Is it used when multiple ap's are in different vlans
-> OR when access points and controller are in different vlan
If both ap's and controllers are in same vlan still the above options have to be configured?

One should be enough 78 is easier to configure. VLAN's should not matter as long as the dhcp scopes have the options for the controllers configured.
VCI are all listed on page 53, you can also find out more on the option 78/43 configurations.

ANS: You should not need to configure DHCP options. Only if the ap's are in a different subnet than the AP registration interface on the controller.

Set the esa port to physical (not virtual, example Bridge at Controller), check off ap registration and management.

OK got it..... One final question not related to the post...... If I use the management port of the identify controller to connect to our lan (say l3 switch) What will be the created topologies interfaces be set to.....say I want to create 10 topologies (B@AP) and only one link of controller (mgmt port) will be used that is connected to l3 switch ....

Option 78 is not on the controller by default, on layer 2 the controllers/aps do L2 SLP, the ap is setup to look for the controller without any special configurations needed. You can use 43/60 on your server if 78 is not available.
