I don't have experience upgrading of VSP 7254 port license.

Is it necessary to reboot after upgrade of VSP 7254 port license ?

I believe it is not needed. You can use the load license CLI command to perform this action.
However, if the loading of the license fails I have seen that the switch performs a reboot, so I would advise to not do this during full production hours.
